Synta Pharmaceuticals is a biopharmaceutical company focused on discovering,
developing, and commercializing novel pharmaceutical products for extending and
enhancing the lives of patients with severe medical conditions, including
cancer and chronic inflammatory disease. Synta has a unique chemical compound
library, an integrated discovery engine, and a pipeline of small molecule drug
candidates with novel chemical structures and distinct mechanisms of action.
All clinical candidates were invented internally by Synta scientists using our
compound library and discovery capabilities. Our lead clinical candidate,
elesclomol, is in a global, pivotal, Phase 3 clinical trial for metastatic
melanoma. Elesclomol is being jointly developed and commercialized with
GlaxoSmithKline; Synta retains all rights, in all indications and markets, to
all of our other drug candidates and programs.
Synta: big pharma assets, small pharma speed

Mechanism diversity and pipeline depth: Synta has three programs in
clinical development, two programs in preclinical development, and additional
programs in discovery. Each program represents a distinct chemical family,
mechanism of action, and therapeutic opportunity. We actively develop second or
third generation compounds for each program to ensure strong IP protection and
competitive positioning.

High quality clinical data: We run double-blind, randomized, controlled,
multi-center Phase 2b and Phase 3 trials whenever practical and appropriate. We
believe the rigorous safety and efficacy data we generate earlier in
development lead to better decision making and resource allocation later in
development, including Phase 3 trials that are confirmatory rather than
exploratory.

Phase 3 clinical program: Our most advanced drug candidate, elesclomol,
is currently enrolling patients in a pivotal, Phase 3 trial in 630 patients,
with product launch targeted for 2009. Elesclomol acts through a novel
mechanism of action, oxidative stress induction, to trigger programmed cell
death in cancer cells. In a double-blind, randomized, controlled study in
patients with stage IV melanoma, elesclomol plus paclitaxel doubled
progression-free survival compared to paclitaxel alone (p=0.035). Elesclomol
has received Fast Track designation from the FDA and completed a Special
Protocol Assessment (SPA) for the pivotal study.

Productive discovery engine and chemistry platform: Our unique chemical
compound library – proprietary compounds collected over two decades from
industrial and academic sources not typically found at pharmaceutical firms –
combined with our high throughput screening and in vivo modeling capabilities;
our medicinal, computational, analytic, and process-development chemistry
expertise; and our in vitro and in vivo toxicology and DMPK experience allow us
to continue to generate promising new drug candidates each year, and set high
standards for those compounds we choose to advance into clinical trials.

Strong financial and strategic position: Our cash position; the financing
provided by our partner for our lead program; and the strategic benefits of
having one partnered and four unpartnered promising, large-market drug
candidates provide a strong financial and strategic foundation for our future.

Strong intellectual property position: We have an extensive intellectual
property portfolio consisting of over 500 issued or pending patents. All Synta
compounds currently in clinical trials represent novel chemical structures
protected by issued or allowed composition of matter patents, as well as a
robust package of related structure, mechanism, formulation, manufacturing, and
administration patents.

Experienced management team: The Synta management team has deep
experience in all stages of drug discovery, development and commercialization.
In addition to developing our current pipeline from discovery through Phase 3,
including initiating and overseeing nearly two dozen clinical trials, the Synta
management team has collectively been involved in the pre-clinical development,
clinical development, and commercial launch of multiple leading drugs.
